An optical method for achieving liquid-liquid phase separation (LLPS) was reported. By using optical tweezers (OT) technique, the highly focused laser trap can produce a microdroplet in the mixture of inorganic phosphate buffered saline (PBS) and organic solvent isopropanol. The Raman test method revealed that the phosphate microdroplet was the product of LLPS. The controllable generation of microdroplets can be achieved by adjusting the proportion of two components in the mixture (1:3 to 1:40). With advantages in precise manipulation and controllable growth of single microdroplet, such an OT-based LLPS method promotes potential innovation ideas in the fields of disease treatment, intracellular structures, and droplet microfluidics.
